The unicellular, halotolerant, green alga, Dunaliella salina has the nique ability to adapt and grow in a wide range of salt conditions. To better understand the molecular basis of its salinity tolerance, a complementary DNA (cDNA) library was constructed from D. salina cells adapted to 2.5M NaCl, salt-shocked at 3.4M NaCl for 5 h, and used to generate an expressed sequence tag (EST) database.
I am part of two major projects:
1. Whole-transcriptome analysis can determine when and where each gene is turned on or off in the cells providing better understanding to biological pathways and molecular mechanisms that regulate disease progression. Individuals with morbid obesity who undergo bariatric surgery represent a unique opportunity to examine the effects of obesity and weight loss on gene expression and our aim is to assess potential changes in gene expression in morbidly obese subjects before and after bariatric surgery. The work will be conducted using Illumina advanced technology HiSeq 2000 instrumentation.
2.Genome-Wide Association (GWAS) study is an approach that involves rapidly scanning markers across the complete sets of DNA, of many people to find genetic variations associated with a particular disease. Once new genetic associations are identified, researchers can use the information to develop better strategies to detect, treat and prevent the disease. Such studies are particularly useful in finding genetic variations that contribute to common, complex diseases, such as obesity, diabetes and other chronic diseases. My contribution to the project is to develop Genome-Wide Association study on Kuwaiti population using Illumina bead-chip array technology starting from participants selection, sorting samples, conducting the experiment and generating the required data files for bioinformatics team.
•Study variant genetic biomarkers SNP's related to Vitamin-D deficiency using Real-Time Taq-Man assay.
•Gene expression analysis for obesity and inflammation related genes using Real-Time Taq-Man assay.
•Member of laboratories committee that is responsible of reviewing protocols and writing standard operating procedures (SOP's) to achieve level of Gold by Accreditation Canada International.
•Train lab personnel on basic and advanced molecular genetic techniques.
•General laboratory organizing issues such as: items ordering, inventory, and generating standard operating procedures to maintain high level of work based on Canadian accreditation standards.
•I involved in several inherited diseases studies to diagnose and discover possible novel genetic cause of the disease. Auto immune deficiency disorders was the major project; I screened candidate genes using Sanger direct sequencing for patients to generate a diagnostic report for known mutations, while patients with negative results were selected for further linkage analysis using Affymatrix genechip protocol.
•Develop new technical assays for mutation detection.
•Manage the research center sequencing core facility using MegaBace DNA sequence instrument and oligonucleotide design and order.
•Train and supervise students for summer interim project, support lab personnel in the requirements for College of American Pathologist accreditation.
•Abiotic stresses have negative impacts on plants; improve the drought, salinity and cold tolerance in grapes (Vitis vinifera) will allow the grapes to be grown in the northern Nevada. The approach was to conduct integrated transcriptome, proteome, and metabolome analysis for different stress conditions; I was involved mainly in applying stresses (drought, salinity, and cold) on plants to develop cDNA libraries from leaves, roots and fruits of V. vinifera. Using molecular biology techniques, I screened about 120,000 EST generated from different libraries to characterize the grape transcriptome.
•Other duties in the lab: organize and follow up orders related to the project, train and supervise undergraduate students to enhance the data mining, care for plants in the green house and the field, and general lab inventory.
