Comparative Genome Analysis and Localization of Hypothetical Conserved Plasmodial Proteins” Research work is divided into two parts, the first part of the study focused on the investigation of genomic differences between parasite isolate causing complicated and uncomplicated malaria from the same geographical location, for this purpose, a tiling array based Comparative Genomic Hybridization approach using 2x400k array Agilent platform. By exploring the genomic variation in the parasite isolate causing complicated malaria in addition to those already reported, we have been able to detect some putative and novel variations. However, the differences in the parasite isolate could also be inherently natural differences between two parasite isolates without any direct correlation with the disease conditions. Another aspect of the study involves cloning, expression and localization of the proteins (hub genes) that were extracted from a co-expression based differentially regulated systems network created from parasite RNA extracted from isolates from patients showing uncomplicated or complicated disease manifestation. For cloning, protein expression and localization studies hub genes from up-regulated module (PFD0300W) and steady-state module (PFL2320W) from the co- expression-based network were selected. The in-silico analysis of the selected proteins suggests the disordered nature of the proteins. Out of the two proteins, localization of one of the proteins (PFD0300W) was successful in the cytoplasm of the parasite in both sexual and asexual stages, of the P. falciparum Indian isolate.
