Dissertation: "Validating Quantifiler Duo vs. Quantiplex HY res kits to Quantifiler Human"
As part of Bay Path University's Forensic Science Graduate Program, in the Forensic Biology Research course I conducted an internal validation study of the Quantifiler Duo kit and Investigator Quantiplex HY res kit to detect low target DNA in biological samples. The validation was conducted in accordance with the FBI's Quality Assurance Standards guidelines for forensic DNA analysis. This study provided an insight in the realm of forensic science for DNA quantitation. PCR quantitation is useful in revealing a person's identity from a case based on the DNA found in biological samples. Based on the results from the Real-time PCR (qPCR) and analysis with Capillary Electrophoresis (CE), electropherograms of STR profiles would be reproduced and compared. The conclusion will be used to provide a recommendation for the implementation of an appropriate kit due to the amplification of DNA. With further study, the validation results will potentially be submitted to Applied Biosystems and Qiagen as a case study.
