Plan on outlining between 10 and 12 questions you want to ask before the day of the interview. These questions should revolve around what the interviewee does in a typical day, what they like about their job, their experience up to that point, what they find most satisfying and challenging about their position, etc.
Don't be afraid to stray from your list of questions if the conversation goes in a particular direction. Most informational interviews are relatively casual, so it is appropriate to ask questions in a conversational manner as they come to mind. Overall, keep the conversation brief, friendly, and focused on the interviewee's personal experiences and career field.
