Professional Summary
I have a broad background in molecular and cell biology, with specific training and expertise in cell adhesion molecules a key research area for my current lab. As a graduate student in Dr. Eric Villard's lab at INSERM UMR 956, Paris, I studied the molecular and cellular mechanisms of the Arrhythmogenic Right Ventricular Cardiomyopathy (ARVC). ARVC is a rare cardiomyopathy associated with fibro-fatty replacement, arrhythmia and sudden death but can also be associated with palmoplantar keratodermia and woolly hair in some case (called Naxos disease and Carvajal syndrome). This pathology is the consequence of mutations in genes encoding desmosomal proteins. My first objective was to identify a specific signature of junctional protein expression on a collection of ARVC heart explants. During this work, I was specifically interested in Desmoglein-2 (DSG2), a desmosomal cadherin, which is dramatically decreased in these patients. Therefore, my second objective was to study the mechanism(s) by which specific DSG2 mutations lead to a severe ARVC phenotype. These mutations have the capability to inhibit the propeptide cleavage, which are associated with a complete loss of interaction in classical cadherins. In this study, I have shown that these DSG2 mutations don't affect the cadherin function in normal culture condition. However, these mutations exhibit a phenotype during mechanical stretching and strongly affect the desmosome structure. I also, as a parallel study, to identify new genetic cause to the disease by  genes candidates approach on a population of ARVD/C probands. During my Ph.D. I was in charge of developing this project, which was a new research area in the laboratory. I acquired experience in growing cardiomyocytes under stress conditions (Hydrogels/mechanical stretch) and a large panel of molecular biology techniques. I joined Dr. Glenn Radice's lab in the Center for Translational Medicine to expand on my previous experience in molecular cardiology with training in mouse molecular genetics, cell biology, and cardiac physiology. I am investigating the role of the cytoskeletal linker proteins a-catenins in normal cardiac development and injury response. I have the expertise and motivation necessary to successfully carry out my knowledge.
