A resume headline is essentially one or two lines that describe who you are at a glance. This may be something like, "Highly motivated sales professional. A resume headline is far from mandatory, but it can be effective at catching employers' attention if it is correctly used.
The resume header, on the other hand, is simply a list of your personal information. The header needs to include your full name, mailing address, phone number, and email address. In a way, the headline is a part of the header. However, the header is a mandatory part of any resume, so be sure to include one in your resume.
